Certain chemicals cause epigenetic changes that foster illness in rats? offspring

Exposure to certain pollutants early in a rat?s pregnancy can foster disease in her offspring during their adulthood as well as in subsequent generations, a new study shows. A wide range of pollutants elicited such lasting effects, despite future generations never encountering the triggering pollutant.

Some chemicals tested led to premature puberty among great-granddaughters, with an increased risk of disease in reproductive tissues. In some tests, the chemicals disrupted ovarian function, leading to infertility or premature menopause. And another chemical exposure caused premature death of sperm-forming cells in the great-grandsons, researchers report online February 28 in PLoS ONE.

Rather than altering genes, the tested pollutants altered chemical switches that regulate genes, reports Michael Skinner and his colleagues at Washington State University in Pullman. These epigenetic switches can lock a gene on or off.

These master switches for DNA are fairly easy to modify throughout life. Early in development, a fetus erases any epigenetic changes acquired during its parents? lifetimes, resetting those switches back to healthy, default programming.

Because the fetus has a mechanism to erase such changes, a pollutant?s epigenetic effects shouldn?t occur in subsequent generations, says epigeneticist John McCarrey of the University of Texas at San Antonio, who was not involved in the study. That they did emerge in the new study ?is pretty heavy in terms of their potential significance,? he says.

In the new work, Skinner worked with the Army, which funded the study, to identify pollutants to which troops would probably be exposed. They settled on dioxins (spewed by burning materials or the defoliant Agent Orange); the insect deterrents DEET and permethrin; the plastic ingredients bisphenol A and phthalates; and jet fuel (often sprayed onto dirt for dust control). ?I tried to pick classes of compounds that were across the whole [pollutant] spectrum ? everything from hydrocarbons down to an endocrine disruptor like bisphenol A,? Skinner says.

The exposures used were relatively high, but not high enough to cause fetal deaths or signs of toxicity in either exposed rat moms or their pups. Yet ?all promoted epigenetic transgenerational changes,? Skinner says. This suggests that epigenetic changes that get passed down through the generations are not some unique quirk of any one chemical, he says; he now suspects most pollutants have the potential to do this.

Each group of pollutants caused a different constellation of epigenetic changes that were linked with various reproductive problems in subsequent generations, although there was some overlap. ?This means we might actually have a means in the future to use epigenetic biomarkers to assess what you ? or your ancestors ? were exposed to earlier in life,? Skinner says, and perhaps apply that knowledge to predict disease and limit risk.

A giant 'little step' in cancer treatment opening up new therapeutic horizons

ScienceDaily (Feb. 27, 2012) ? A study headed up by the Vall d?Hebron Institute of Oncology (VHIO) heralds a new horizon in the fight against cancer, opening up a parallel dimension to existing treatment options. The data, published today in the Journal of Clinical Oncology, demonstrate that by combining two drugs that had already been used on a patient in the past but had stopped working, they boost each other?s efficacy and at the same time manage to break down the patient?s resistance to each of them individually, presenting a third potential treatment option for clinically advanced metastatic tumours. While this may not initially appear to be an innovation, given that combined treatments are used in cancer as a matter of course, it actually represents a radical change in the use of the existing therapeutic weaponry, an extremely practical solution and a source of hope for many patients.

?The novelty factor does not lie in the combination itself, but in the combination of two drugs that had already been administered in the past and had each failed,? explained Dr Javier Cort?s, the Head of the Breast Cancer Programme at the VHIO and main author of this work. This not only offers new alternatives for patients with advanced tumours ?the most important aspect ? but also opens up a parallel dimension of new therapeutic options to the existing treatments, given that it offers numerous combinations between existing pharmacological treatments that not only boost each other but also counteract patients? resistance to them.

?The study focused on HER2 positive breast cancer, although the concept could be applied to the treatment of other tumours?, continued Dr Cort?s. Breast tumours occur most frequently in Western women. In Spain alone, some 24,000 new cases are diagnosed each year, and it is calculated that one in every eight or nine women in this country will suffer from breast cancer. Although the many advances over recent years in screening, early diagnosis and treatments have dramatically reduced mortality from this disease, there are still breast tumours whose biological characteristics make them more aggressive.

Around 15% of breast tumours are HER2 positive and fall into this category. Moreover, they tend to affect younger women. In spite of the fact that this group has benefited tremendously from specific biological therapies targeted at this subtype (monoclonal antibodies), many tumours continue to advance and metastasize. ?When a patient with this breast cancer subtype stops responding to the initial drug and the disease progresses, a second drug is administered. It may be that the response to these two treatments is negligible or there may even be no response at all and the tumour continues to grow.If we then treat the patients with the two drugs in combination, even though they ceased to function individually, what we have found is that almost 50% of the tumours responded to this combination and did so very positively, showing a clear improvement or keeping the disease stable for a considerable period of time?, concluded Dr Cort?s.

In the fight against cancer, the combination of treatments is highly common practice. However, the concept described is different: it is not about seeing the results of combining drug A with drug B, but, when A fails to work, B is administered, and if this also fails we return to the starting point and initiate treatment with a combination of the two (A + B).

This strategy could offer an unprecedented key to multiplying treatment options. In the specific case of this study, we administered trastuzumab (the first-choice treatment for HER2 positive breast cancer); when the tumour continued growing we used pertuzumab (currently pending approval) and when the cancer continued to progress we administered the two together. ?The results were very positive and highly informative for new studies,? said Dr Cort?s.

NEW YORK (CNNMoney) -- Expect the upward march of oil and gas prices to overshadow bellwether corporate earnings, economic reports and a read on the health of the European banking sector -- all of which are due next week.

Investors will spend most of next week grappling with whether the rising price of oil could crimp the stock market's 2012 rally.

All three indexes eked out another week of roughly 0.5% gains. The Dow Jones industrial average (INDU) is up 6.3% so far this year, hitting its highest level since May 2008. For 2012, the S&P 500 (SPX) is up 8.6%, and the Nasdaq is up 13.8%.

Still, economists and analysts worry that rising gas prices could cause consumers to cut back on spending, which could threaten an already shaky economic recovery.

U.S. crude oil prices are at their highest levels since May 2008, hitting $109.77 a barrel. U.S. oil prices climbed nearly 6% this week. In turn, the average price of a gallon of gas jumped more than 13 cents..

Investors will watch to see what Federal Reserve Chairman Ben Bernanke says about energy prices and the broader economy when he delivers his semiannual address to Congress on Wednesday.

"If there are any signs that the Fed would be supporting the Treasury or mortgage markets later this year because of energy prices, investors could take that as a buying opportunity," said Robert Tipp, chief investment strategist at Prudential Fixed Income.

The other key economic reports on tap this week are the Case-Shiller Home Price Index for December, due out Tuesday; readings on January's personal income and spending, due out Thursday; and the ISM manufacturing index, also to be released Thursday.

Better-than-expected economic reports have helped fuel the stock market's rally this year, so analysts say investors have largely priced in positive data. Anything that falls below expectations could cause an outsized sell off in stocks.

"There's not a lot of tolerance for bad numbers," said Uri Landesman, president of the fund Platinum Partners. "The market is discounting good news."

Meanwhile, company earnings are playing a larger role in the overall market and in driving prices of individual stocks. That's a sharp turnaround from 2011, when stocks largely moved in correlation with each other based on broader economic news.

"Greece is appearing to be moving into the rearview mirror, and stock-specific stories are coming into view," said Dan Greenhaus.

Earnings season is nearing a close, yet numerous bellwether stocks will report quarterly results this week: Home improvement retailer Lowe's (LOW, Fortune 500) and Priceline (PCLN) report earnings Monday. AutoZone (AZO, Fortune 500), Cablevision (CVC, Fortune 500) and Dreamworks Animation (DWA) report results on Tuesday. Retailer Liz Claiborne, where shares have jumped 20% in 2012, will report Wednesday. Jeremy Lin's corporate parent Madison Square Garden (MSG) will release results Friday.

The threat of Europe's sovereign debt crisis is taking a momentary backseat to economic and corporate news, but investors will spend Wednesday digging into how much debt eurozone banks will take from the European Central Bank.

The central bank will hold its second auction to allow banks to borrow three-year loans at low interest rates through its so-called long-term refinancing operation (LTRO) program. In its first auction in December, banks borrowed ?489 billion from the ECB. Analysts expect that number to increase this time around.

"This program requires a very nuanced read," said Prudential's Tipp. "It will take some time for analysts to disentangle the numbers, but it gives us a window into the European banking system to see how desperate different institutions are." ?To top of page

iPad 3 Shipments Have Begun as Release Date Nears: Report

Foxconn has started shipping the highly anticipated iPad 3 ahead of Apple's official unveiling of the tablet in March, according to MacRumors, which came across a shipping document on a Chinese Web site.

The document on the Twitter-like Web site Sina Weibo shows details of shipments beginning Sunday from Apple supplier Foxconn to the United States.

A translation of the document by MacRumors indicates that the tablet will ship into three major U.S. airports with maximum cargo security and initial deliveries will begin March 9.? MacRumors has also released a picture of the tablet being uploaded onto a plane.

With just weeks left until Apple's anticipated official unveiling of the iPad 3, a Chinese blog claims to have leaked pictures of the next-generation tablet.

The?Apple Daily?has published unconfirmed pictures of an iPad 3 that looks much like the iPad 2. The pictures show the tapered back of the tablet and parts of its apparent inner mechanisms.

The?Chinese blog?claims to have discovered an Apple A6 chip, which is the prime rumored processor of the iPad 3. It also says the new iPad will come with 4G connectivity as opposed to 3G found in the iPad 2.

The iPad 3 will come with a 9.7-inch Retina display, like the iPad 2, with a 2048?1536 resolution, according to the blog, which matches popular rumors.

The pictures?show a camera, which the blog claims is an 8-megapixel camera to match the iPhone 4S and better the 0.8MP camera of the iPad 2.

If difficult to tell if the photos are legitimate, but all will be unveiled by Apple on March.7. In the meantime here is a roundup of?the latest rumors:

Improved Display:

The iPad 2 has been heavily criticized for its 1024x768 resolution with owners complaining about unclear fuzzy images. The iPad 3 will undoubtedly come with a better display.?It's rumored to quadruple its pixel destiny, but it's most likely to just double that of its predecessor, according to SlashGear.

The DigiTimes, who claim to have received information from Apple's supply chain, say the new display will boast an HD display panel with a resolution of 2048x1536 pixels.

Faster Processor:

The iPad 3 will likely come with a more powerful chip, perhaps the quad-core A6 chip. The first-generation iPad came with an A4 Chip and the iPad 2 followed with an A5 chip, which suggests the iPad 3 will follow suit and run on the A6 chip.

An A6 chip would double the power of the A5 chip that is currently used in the iPhone 4s and the iPad 2. An unnamed source, who claims to have an iPad 3 prototype, sent?Boy Genius Report?pictures of a diagnostic tool called iBoot, which states a chip model number that appears to be that of the A6 chip.

Better Camera:

The iPad 2's camera is another feature of the tablet that has come under strong critique, sporting a feeble 0.8MP. The iPad 3 has a long way to go if it wants to compete with other tablets like the ASUS Transformer Prime, which currently boasts an 8MP, 3264 x2448 pixel, auto-focus, LED flash camera.

The iPad 3 will likely come with a much-improved camera, matching the quality of the 8MP camera of the iPhone 4s or better. The iPhone 4S not only improved its pixel capacity by 60 percent compared with its predecessor, but it also came with a larger custom lens that heralded a 2.4 aperture. More, the camera boasts advanced color accuracy, face detection and reduced motion blur.

Price:

While rumors continue to circulate about the possibility of two iPad 3 models -- one cheaper than the other -- it is likely there will only be one, which will fall under the same price range of its predecessor at $499 for its entry level model. The iPad 2 will likely drop to $399 and if Apple continues to sell the original iPad it will probably start at $199.

Mark Post, a physiologist at the University of Maastricht in the Netherlands, said the project is being funded by an anonymous investor who is interested in "life-transforming technologies" and believes lab-grown meat could revolutionize the food industry.


"It's a reputable source of money, I can tell you," Post said today in Vancouver, Canada, at the annual meeting of the American Association for the Advancement of Science.

Post hopes the tasting will be a media event, with experimental chef Heston Blumenthal cooking the burger. The patty will be much like a regular quarter-pounder?? but with one big difference: This one will be created by growing bovine stem cells in a vat, transforming them into thousands of thin layers of beef muscle cells, mincing them into tiny pieces, then combining the bits with lab-grown animal fat to form a lump of meat the size of a golf ball.

If Post and his colleagues succeed, it would mark a technological triumph after years of working to improve upon the current, millennia-old method for making meat. Researchers in the field say the livestock industry in its current incarnation is too energy-intensive and land-intensive for a global population that's rising in numbers and affluence.

Meat production already takes up more than half of the world's estimated agricultural capacity, in one way or another. U.N. figures show that animal farming takes up 30 percent of the planet's exposed land mass. And over the next 40 years, the demand for meat products is expected to double.

If the researchers' assumptions are correct, growing meat in the lab "could reduce the energy expenditure by about 40 percent," Post said. Lab-grown meat has also won the endorsement of People for the Ethical Treatment of Animals, or PETA, because the stem cells could be extracted without killing animals.

The money behind the meat
Post has been talking about serving up the first lab-grown burger for a long time, but it took the anonymous 250,000-euro ($330,000) contribution to turn the dream into reality. Traditional meat producers weren't interested in changing their ways, and were doubtful about success, he said. "Most people don't believe it's ever going to happen," he told reporters.

When Post started working on the project, he focused on growing stem cells from pigs to create a lab-grown sausage, but he said "my financier was not very interested in sausages."

There's still a long way to go between now and the celebrity cookout: Post said he doesn't yet know what the burger would taste like, because the samples that have been grown so far are too small. The pinkish-yellowish strips of muscle cells are only about an inch (3 centimeters) long, a half-inch (1.5 centimeter) wide, and so thin (1 millimeter) that they're semi-transparent. Post feels confident that his team can perfect the process by October, but full commercialization could take another 10 years or more.

The good news is that if there's someone out there willing to buy the second lab-grown hamburger, they can get it for "an extreme reduction in price," Post told me. He estimates that piece of meat should cost just 200,000 euros ($263,000).

Beyond meat
It's worth asking whether the quest to grow lab-grown meat is worth the effort, considering that there are already vegetarian alternatives to meat. Aren't tofurky and field roast good enough? Post and others note that such products haven't made a significant dent in the meat market, and are generally more expensive than the meat items they're meant to replace.

"If there is a vegetable-derived product that can take away the human being's craving for meat, that would be preferable," Post said.

Stanford University biochemist Patrick Brown says he's working on precisely that kind of stuff, and it could be on the market in the next year or so.

"We have a class of products that just totally rocks and cannot be distinguished from the animal-based product it replaces, even by hardcore foodies," he said. He promised that his plant-based alternatives to meat and dairy products would be tasty, nutritious ? and profitable.

"I think it's going to be one of the easier things I've done," he said.

Brown joked that he couldn't talk about the details, "because if I did, I'd have to kill you." He'd say only that he "had no trouble getting investment" from a Silicon Valley venture-capital firm. To commercialize the concept, two ventures have been set up with placeholder names: Sand Hill Foods and Jasper Ridge Creamery.

The way Brown sees it, the meat industry is a "sitting duck for disruptive technology," offering a rich target for alternatives. He said the wholesale market for unprocessed meat has been estimated at $150 billion a year, which is 250 times the current market for meat alternatives.

Even though Post said the meat industry has been generally standoffish about lab alternatives, some companies are going against the grain: Nicholas Genovese, a visiting scholar at the University of Missouri at Columbia, told journalists that JBS, one of the world's biggest meat-packing companies, was interested in his parallel effort to grow meat in the lab.

Gamers Pave The Way For Cheap Cancer Research | WebProNews

Gamers are somewhat reviled by the media and politicians as being violent psychopaths who use violent first-person shooters and other ?murder simulators? to play out acts that may lead to real life consequences. Those extreme graphics causing all the violence, however, may end up saving lives.

A research lab at Wake Forest University is using graphic processing units to simulate the inner workings of human cells according to ScienceBlog. Samuel Cho, biophysicist and computer scientist says that the use of the GPUs is furthering their research by leaps and bounds.

?If it wasn?t for gamers who kept buying these GPUs, the prices wouldn?t have dropped, and we couldn?t have used them for science,? Cho says.

The team uses the GPUs to see how cells live, divide and die. This will help them create targets for tumor-killing drugs.

Cho recently created a simulation that shows the folding of a critical RNA molecule that?s part of the human telomerase enzyme. It gives scientists a better look at how these cells operate, including the revelation of hidden states in the folding and unfolding process.

That big word up there, the telomerase enzyme, it?s only found in cancerous cells. It essentially keeps cancer cells from dying.

?The cell keeps reproducing over and over, and that?s the very definition of cancer,? Cho says. ?By knowing how telomerase folds and functions, we provide a new area for researching cancer treatments.?

With their research, they should be able to create a drug that stops the reproduction of this molecule so the cancer cell dies.

Now back to the gaming bit, they are using GPUs to research the bacterial ribosome ? a molecular system 200 times larger than the telomerase molecule. They are using the GPUs to simulate the cell?s growth at a much faster rate.

Cho estimates that this research would have taken them over 40 years to complete with traditional cell imaging. With the use of GPUs, it will only take them a few months.
